STEM-27. MECHANOSENSITIVE BRAIN TUMOR CELLS CONSTRUCT BLOOD-TUMOR BARRIER TO MASK CHEMOSENSITIVITY

چکیده

Abstract Two major obstacles in brain cancer treatment are the blood-tumor barrier (BTB) and quiescent tumor cells. Here we show that Sox2+ cells directly project cellular processes to ensheathe capillaries medulloblastoma (MB), a process depends on mechanosensitive ion channel Piezo2. MB develops tissue stiffness gradient as function of distance capillaries. perceive substrate sustain local intracellular calcium, actomyosin tension, adhesion promote growth cell surface sequestration β-Catenin. Piezo2 knockout reverses WNT/β-Catenin signaling states between endothelial cells, compromises BTB, reduces quiescence markedly enhances response chemotherapy. Our study reveals construct BTB mask chemosensitivity. Targeting addresses properties underlie therapy failures cancer.

An overview of therapeutic approaches to brain tumor stem cells

Primary and secondary malignant central nervous system (CNS) tumors are devastating invasive tumors able to give rise to many kinds of differentiated tumor cells. Glioblastoma multiform (GBM), is the most malignant brain tumor, in which its growth and persistence depend on cancer stem cells with enhanced DNA damage repair program that also induces recurrence and resists current chemo- and radi...

Brain Tumor Stem Cells and Immunotherapy

Glioblastoma multiforme (GBM) is one of the most common aggressive primary brain tumors, and it responds poorly to the current treatment combination of surgery, radio-, and chemotherapy. The hypothesis that cancer stem cells may account for the pathogenesis underlying various tumors, including GBM, has been accepted widely in recent years. Brain tumor stem cells (BTSCs) have been shown to contr...

Brain Tumor Stem Cells Remain in Play.

Traditional anticancer therapy includes the administration of harsh chemicals, often coupled with targeted radiation therapy. This strategy often delivers positive results in terms of reduced tumor burden, but the effects are rarely lasting and all too frequently the disease progresses. In addition, chemoand radiotherapy do not discriminate between normal cells and cancer cells, but rather targ...

Molecular Culprits Generating Brain Tumor Stem Cells

Despite current advances in multimodality therapies, such as surgery, radiotherapy, and chemotherapy, the outcome for patients with high-grade glioma remains fatal. Understanding how glioma cells resist various therapies may provide opportunities for developing new therapies. Accumulating evidence suggests that the main obstacle for successfully treating high-grade glioma is the existence of br...
